Research Overview
Epitalon vs pinealon compares two short synthetic peptides from one program: the peptide bioregulator work of V. Kh. Khavinson's group at the St Petersburg Institute of Bioregulation and Gerontology. Epitalon, the tetrapeptide Ala-Glu-Asp-Gly, was designed from the amino acid makeup of epithalamin, a bovine pineal extract, and later detected in a pineal polypeptide complex (Araj et al., 2025). Pinealon is the tripeptide Glu-Asp-Arg. The two share a Glu-Asp core, and papers from the program have tested them side by side (Fedoreyeva et al., 2011).
Epitalon papers center on telomerase in cultured cells and on pineal endpoints such as melatonin. Pinealon papers center on oxidative stress and neuron shape in cell culture. Disguised Alpha supplies Epitalon as lyophilized powder in 10MG and 50MG vials and Pinealon as a 20MG lyophilized vial, for laboratory research use only. Each also has a second format: Capsulated Epitalon and a pre-mixed Pinealon Aerosol Spray.
Structure and Class
Epitalon is Ala-Glu-Asp-Gly (AEDG): C14H22N4O9, 390.35 g/mol (PubChem CID 219042). Pinealon is Glu-Asp-Arg (EDR): C15H26N6O8, 418.4 g/mol (PubChem CID 10273502). The product pages and PubChem agree on every value.
Glu-Asp sits at positions 2 and 3 of epitalon and 1 and 2 of pinealon. Epitalon flanks it with alanine and glycine; pinealon adds arginine, the main chemical difference. Epitalon has two acidic side chains and no basic one. Pinealon has the same two acidic side chains plus arginine's basic guanidino group, so it carries less net negative charge at neutral pH. Related program peptides overlap too: cartalax (Ala-Glu-Asp) is epitalon without its glycine, and vesugen (Lys-Glu-Asp) carries the same Glu-Asp pair.
Pinealon is about 7 percent heavier per molecule, so one milligram holds about 2.56 µmol of epitalon but about 2.39 µmol of pinealon as free peptides. Structural work is thin: an outside review calls physicochemical and structural studies of epitalon limited (Araj et al., 2025).

Research Context: Epitalon
The best-known epitalon claim is telomerase activation. The first report added the peptide to telomerase-negative human fetal fibroblast cultures and described catalytic subunit expression, telomerase activity and telomere elongation (Khavinson et al., 2003). In 2025, a Brunel University London group outside the program tested the breast cancer lines 21NT and BT474 plus normal epithelial cells and fibroblasts.
In the normal cells, telomere length rose with concentration through hTERT and telomerase; in the cancer lines, it rose through alternative lengthening of telomeres (Al-Dulaimi et al., 2025). That paper carries a published correction (PMID 41240216). Both are cell-culture findings, and no controlled human trial has tested the question.
Pineal endpoints are mixed. In old female rhesus monkeys, the originating group reported higher evening melatonin and described the daily cortisol rhythm as normalized (Khavinson et al., 2001). A perifusion study of pineal glands from young and old Wistar rats found no significant effect on melatonin secretion at 1 to 100 µM, or on the response to the beta-adrenergic agonist isoproterenol (Djeridane et al., 2003). That negative study ran in Paris, with Khavinson and V. N. Anisimov as co-authors.
Roughly 160 PubMed records mention epitalon, and about two thirds list Khavinson or Anisimov as an author, many in Russian-language journals. The only record typed as a controlled clinical trial comes from that group too (Khavinson et al., 2002).
Research Context: Pinealon
The most direct pinealon work put cerebellar granule cells, neutrophils and PC12 pheochromocytoma cells under oxidative stress. The peptide limited reactive oxygen species build-up in a concentration-dependent way, cut necrotic cell death, delayed ERK 1/2 activation and changed the cell cycle. Because the cell-cycle change continued above the concentrations that saturated the oxidative effect, the authors proposed a second action at the level of the genome (Khavinson et al., 2011).
The clearest outside work is physical chemistry. A St Petersburg State University physics group, with no Khavinson co-author, used spectroscopy, nuclear magnetic resonance, viscosimetry and molecular dynamics. It reported that pinealon partly enters the major groove of DNA, contacting guanine N7 and O6, and that magnesium ions promote the binding by screening the phosphate charges (Silanteva et al., 2019).
The record is small. A PubMed search for pinealon and its EDR name returns fewer than 50 records, several of them false hits, and most relevant ones come from Khavinson's group or labs working with the same institute. No pinealon record is typed as a controlled clinical trial, and the group's own review calls the model of cell entry and binding to histones or RNA an assumption (Khavinson et al., 2020).
Choosing Between Them for a Research Question
At least six papers from the program and its collaborators tested both in one design; three are summarized here. In organotypic pineal culture from 3-month-old rats, epitalon raised expression of the proliferation marker Ki-67 and CGRP synthesis by pinealocytes, pinealon raised neither, and none of the four peptides tested changed the apoptosis marker AIF (Khavinson et al., 2011).
In induced cortical neurons made from elderly donors' skin fibroblasts, both increased primary dendrite number and total dendrite length, but only pinealon was reported to lower oxidative DNA damage (Kraskovskaya et al., 2024). In HeLa cells, fluorescein-labeled forms of both reached the nucleus and nucleolus, and in oligonucleotide assays both favored CNG-containing sequences, CAG in particular (Fedoreyeva et al., 2011).
So pineal tissue questions, such as pinealocyte proliferation and secretory markers, and telomerase questions point to epitalon. Oxidative stress in neuronal or PC12 cultures, oxidative DNA damage in neurons, and peptide-DNA binding chemistry point to pinealon. Dendrite readouts moved with both, so they cannot separate the two on their own.
Neither is a drop-in substitute for the other. Compare them on moles, not milligrams, and name each by sequence in methods, since papers mix trade names and letter codes.
Handling and Storage
Both vial products ship as lyophilized powder, and both product pages list the same storage: -20°C for the powder as shipped, 2 to 8°C after reconstitution, protected from light, with freeze/thaw cycles avoided. Let a sealed vial reach room temperature before opening, and split stock into single-use aliquots.
Both peptides dissolve readily in water. Epitalon has no basic side chain to offset its two acidic ones, so an unbuffered solution can sit well below neutral; record the pH. In DNA-binding work with pinealon, magnesium promoted the binding (Silanteva et al., 2019), so hold buffer ions constant across comparisons.
The second formats store differently, as listed on their product pages. The Pinealon Aerosol Spray arrives pre-mixed: 15 to 25°C unopened, never frozen, then 2 to 8°C once opened, upright, closed and away from light. Capsulated Epitalon ships as sealed capsules kept at 15 to 25°C, away from moisture and direct light. For any format, record the lot number and check identity and content against its certificate in the COA portal.
Questions Researchers Ask
Epitalon vs pinealon: what is the main difference?
Sequence and research focus. Epitalon is the tetrapeptide Ala-Glu-Asp-Gly, studied mostly for telomerase in cultured cells and for pineal endpoints (Khavinson et al., 2003; Djeridane et al., 2003). Pinealon is the tripeptide Glu-Asp-Arg, studied mostly in oxidative-stress and neuron culture models (Khavinson et al., 2011).
Epithalon vs pinealon: is epithalon a different peptide?
No. Epithalon and epithalone are other spellings of epitalon, the same Ala-Glu-Asp-Gly peptide (PubChem CID 219042). Epithalamin is different: it is the bovine pineal extract the tetrapeptide was designed from (Araj et al., 2025). Pinealon is a separate three-residue peptide.
Pinealon vs epitalon: which has the stronger evidence?
Neither is strong by usual standards. Epitalon has the larger record and an outside telomere study that carries a published correction (Al-Dulaimi et al., 2025). Pinealon has fewer than 50 records and one main outside study, on DNA binding (Silanteva et al., 2019). For both, most work comes from one institute and uses cell culture or animals.
Do they share a mechanism?
One is proposed, not established. The originating group suggests that short peptides enter the nucleus and bind DNA by sequence (Fedoreyeva et al., 2011). Its own pinealon review calls cell entry and binding to histones or RNA an assumption (Khavinson et al., 2020), and an outside epitalon review says it is uncertain whether the known actions are its only mechanisms (Araj et al., 2025).
Epitalon and Pinealon are supplied strictly for laboratory and in-vitro research use. They are not for human consumption, veterinary use, or any diagnostic or therapeutic application. Nothing on this page is guidance for use in people.
