P21 is a small synthetic peptide mimetic built from ciliary neurotrophic factor, or CNTF. Papers also call it P021 or Peptide 021. It was designed in Khalid Iqbal's lab at the New York State Institute for Basic Research. We supply P-21 as a 10MG lyophilized vial.

Our store lists it as P21 (BDNF), but P21 is not BDNF. BDNF is a much larger natural protein. P21 is studied for its effect on BDNF expression in rodent brain.

What the research covers

The first paper wrote P21 as Ac-DGGLAG-NH2 (Li et al., 2010). Its four amino acids, Asp-Gly-Gly-Leu, come from human CNTF. An adamantylated glycine sits at the end. Its developers propose that it acts by blocking leukemia inhibitory factor (LIF) signaling and raising BDNF expression (Kazim and Iqbal, 2016).

All published P021 work is in mice, rats and cell cultures. Every P021 study in PubMed lists Iqbal as an author, so outside replication is thin.

  • Neurogenesis in the dentate gyrus.
  • BDNF expression in rodent brain.
  • Tau and amyloid measures in transgenic mice.
  • Synaptic plasticity markers.

That 2010 study in normal adult mice reported more new neurons in the dentate gyrus, and more of them maturing. A 2014 study in rats aged 22 to 24 months reported higher BDNF expression (Bolognin et al., 2014). In 3xTg-AD mice, a 2014 study reported less abnormal tau hyperphosphorylation and lower soluble amyloid beta (Kazim et al., 2014). A 2024 study in Cdkl5 knockout mice found no rise in brain BDNF (Mottolese et al., 2024).
