Research Overview

NMN and NR sit one step apart in the NAD+ salvage pathway. NR (nicotinamide riboside) is a nucleoside, nicotinamide bonded to ribose; NMN (nicotinamide mononucleotide) is the same unit with a phosphate on the ribose. Nicotinamide riboside kinases, first described in yeast and humans in 2004, add that phosphate (Bieganowski and Brenner, 2004). NMN adenylyltransferases (NMNATs) then join NMN to ATP to make NAD+ (Nikiforov et al., 2011).

So the difference between NMN and NR is one phosphate and one enzyme step, and that step drives every other contrast in an NMN vs NR comparison. Disguised Alpha supplies NMN as 500MG capsules and NAD+ as buffered lyophilized vials and a 200MG/ML liquid, for laboratory research use only. It does not list nicotinamide riboside, so the NR data here are literature reference. Nothing on this page is guidance for use in people.

Structure and Class

Both are pyridine compounds built on the same nicotinamide and ribose unit; neither is a peptide. NMN is a zwitterionic nucleotide: C11H15N2O8P, 334.22 g/mol (PubChem CID 14180), as on the product page. NR is a cation, C11H15N2O5+, 255.25 g/mol (PubChem CID 439924), so it exists as a salt; the chloride used in the first human pharmacokinetic study (Trammell et al., 2016) is C11H15ClN2O5, 290.70 g/mol (PubChem CID 90480033).

The kinases are NRK1 and NRK2, which phosphorylate NR to NMN (Bieganowski and Brenner, 2004). In mammalian cells, including mouse hepatocytes, NRK1 was necessary and rate-limiting for NAD+ synthesis from added NR and from added NMN (Ratajczak et al., 2016).

Research Context: NMN

How NMN enters cells is the most argued point here. In mouse primary hepatocytes, NR and NMN raised NAD+ only in cells with NRK1. Doubly labeled NR labeled 40 to 50 percent of cellular NMN and NAD+ within 6 hours, while doubly labeled NMN produced no labeled NMN inside, and the authors concluded that NMN becomes NR outside the cell before uptake (Ratajczak et al., 2016). In serum-free HEK293 cultures with NAMPT blocked, nucleoside transport blockers suppressed NAD+ rescue by NMN as well as by NR (Kulikova et al., 2019).

A 2019 study proposed a direct route: Slc12a8, abundant in mouse small intestine, as a sodium-dependent transporter for NMN but not NR. Knockdown blocked NMN uptake in cells and mice, and Slc12a8 loss lowered NAD+ in jejunum and ileum (Grozio et al., 2019).

A critique said the analytical methods, transport data and interpretation did not support NMN transport (Schmidt and Brenner, 2019); the authors replied with proteoliposome and isotope-tracing data showing, in their reading, intact NMN uptake (Grozio et al., 2019). Both sides disclosed commercial ties: the critique’s senior author to a company that supplies NR, and two original authors to a licensed transporter patent. The question is not settled.

In mice, a 300 mg/kg oral gavage of NMN raised plasma NMN within 2.5 minutes and liver NAD+ from 15 to 30 minutes (Mills et al., 2016). An isotope-tracing study saw little intact NMN: it was barely detectable in mouse blood even after intravenous delivery, which raised circulating NR, and oral NR and NMN were both metabolized to nicotinamide in the liver (Liu et al., 2018). The NMN research guide covers the human trials and US regulatory history.

Research Context: Nicotinamide Riboside

In HEK293 cells, the transporters ENT1, ENT2 and ENT4 imported NR, the concentrative transporters CNT1 to CNT3 did not, and the ENT blocker NBTI stopped NAD+ synthesis from NR; once inside, NR was readily converted to nicotinamide (Kropotov et al., 2021).

In 128 mice given 185 mg/kg NR by oral gavage, or equimolar nicotinamide or nicotinic acid, NR raised liver NAD+ the most. It also raised NAAD, which the authors proposed as a sensitive marker of NAD+ repletion (Trammell et al., 2016). Given intraperitoneally at 500 mg/kg, NR increased mouse liver NAD+ by about 220 percent at 1 hour, against about 170 percent for NMN, and both responses were blunted, not abolished, in NRK1 knockout mice (Ratajczak et al., 2016).

The first clinical pharmacokinetic trial of NR, preceded in the same report by a one-person pilot, was a randomized crossover study in 12 healthy adults, in which three single oral amounts gave amount-dependent rises in the blood NAD+ metabolome; an NR supplier sponsored it (Trammell et al., 2016). A later trial, which its authors describe as the first direct comparison in people, randomized 67 healthy adults to NR, NMN, nicotinamide or placebo for 14 days, open label, and analyzed 65.

Whole-blood NAD+ roughly doubled with NR and with NMN, by comparable margins, but not with nicotinamide. Ex vivo work pointed to gut microbes turning both into nicotinic acid, so the authors proposed a partly gut-dependent route through the Preiss-Handler pathway. All authors were company employees (Christen et al., 2026).

Neither compound is an approved US drug: Drugs@FDA lists no NMN or NR product. FDA’s August 2016 response to GRAS notice GRN 000635 on NR chloride said the agency had no questions at that time about the notifier’s conclusion for its use as a vitamin B3 source in specific foods, and had not made its own determination.

Choosing Between Them for a Research Question

The NR vs NMN choice follows from the step under study, and most mechanism data come from cell lines and mice. Kinase and nucleoside questions point to NR: NRK activity, ENT transport and cleavage to nicotinamide. Nucleotide questions point to NMN: NMNAT activity, Slc12a8 transport and extracellular processing. Test any claim of intact NMN uptake against NR, with NRK1 or nucleoside transport blocked (Ratajczak et al., 2016; Kulikova et al., 2019).

Compare on moles: per milligram, NR chloride carries about 15 percent more molecules than NMN. Concentration matters too: in serum-free HEK293 cultures with NAMPT blocked, 10 µM NR restored metabolic activity to about 80 percent of control and 10 µM NMN did not, while both worked at 100 µM (Kulikova et al., 2019).

Track NMN, NR, nicotinamide and NAD+ in medium and cells, and add nicotinic acid and NAAD for animal or human samples (Christen et al., 2026). Freeze blood samples at once: both compounds had a half-life of about 3 minutes in mouse whole blood (Liu et al., 2018).

Handling and Storage

NMN ships as sealed capsules, 500MG each and 60 per bottle, listed for 15 to 25°C in the original sealed container, away from moisture and direct light. In solution it is the steadier compound: 93 to 99 percent stayed intact in drinking water at room temperature for 7 to 10 days (Mills et al., 2016).

NR hydrolyzes to nicotinamide and ribose in water (Kulikova et al., 2019), so prepare NR solutions fresh, keep them cold, and check by LC-MS what remains at the end of an experiment.

Media differ: fetal bovine serum also broke down NMN (Kulikova et al., 2019), and another lab measured a half-life of about 12 hours for both in medium with dialyzed serum (Liu et al., 2018), so report the medium. For NMN molar work, use the content measured on the lot certificate, and file the certificate from the COA portal with the lot number.

Questions Researchers Ask

What is the difference between NMN and NR?

One phosphate. NR is the nucleoside; NMN is the nucleotide that NRK1 or NRK2 makes from it (Bieganowski and Brenner, 2004), and NMNAT enzymes convert NMN to NAD+ (Nikiforov et al., 2011).

NMN vs nicotinamide riboside: which enters cells intact?

NR does, through ENT1, ENT2 and ENT4 in human cells (Kropotov et al., 2021). For NMN the evidence conflicts: hepatocyte isotope work found it must become NR first (Ratajczak et al., 2016), while the Slc12a8 study reported direct import in mouse intestine (Grozio et al., 2019), which others dispute (Schmidt and Brenner, 2019).

Nicotinamide riboside vs NMN: has a human trial compared them?

Yes, as research history. In a 14-day open-label trial with 65 adults analyzed, whole-blood NAD+ rose comparably, roughly twofold, with each, and did not rise with nicotinamide (Christen et al., 2026).

Is NR more stable than NMN in solution?

No. In culture medium at 37°C, about 35 to 40 percent of NR split to nicotinamide within 24 hours, with or without serum, against about 5 percent of NMN without serum (Kulikova et al., 2019).
