Research Overview

AOD-9604 vs tesamorelin compares two different classes. AOD-9604 is an analog of a fragment of growth hormone (GH) itself, hGH 177-191 with an extra N-terminal tyrosine (Cox et al., 2015), developed to keep GH's lipolytic and antilipogenic signaling without classical GH receptor growth effects or IGF-1 exposure (Dominikowski et al., 2026). Tesamorelin is a stabilized analog of growth hormone-releasing hormone (GHRH), the hypothalamic peptide that signals the pituitary to release GH (Ferdinandi et al., 2007).

They work at different levels: GHRH analogs act upstream at the pituitary GHRH receptor, raising GH and then IGF-1 (Dominikowski et al., 2026), while AOD-9604 was studied for lipolysis and fat oxidation in obese mice and did not compete for the human GH receptor in transfected cells (Heffernan et al., 2001). Disguised Alpha supplies AOD-9604 (current lot labeled 5 mg), tesamorelin (listed in 5MG and 10MG) and a Tesamorelin & Ipamorelin blend (8MG/2MG) as lyophilized powders for laboratory research use only.

Structure and Class

AOD-9604 (also written AOD9604 or AOD 9604) is the 16-residue peptide YLRIVQCRSVEGSCGF, closed by a cysteine 7 to 14 disulfide (PubChem CID 71300630). Analytical work describes it as hGH 177-191 plus an N-terminal tyrosine (Cox et al., 2015); its 16-residue span is why it is sometimes labeled hGH 176-191 (Dominikowski et al., 2026), as on the product page. Product page identity: C78H123N23O23S2, 1815.07 g/mol.

Tesamorelin keeps all 44 residues of human GHRH, YADAIFTNSYRKVLGQLSARKLLQDIMSRQQGESNQERGARARL, with an amidated C-terminus (PubChem CID 16137828), and adds a trans-3-hexenoyl group to tyrosine 1. Developed as TH9507, it resisted dipeptidyl aminopeptidase-IV and broke down more slowly in vitro than native GHRH in rat, dog and human plasma (Ferdinandi et al., 2007). Product page identity: C221H366N72O67S, 5135.86 g/mol.

Research Context: AOD-9604

The isolated adipose tissue work used the parent sequence, hGH 177-191, called AOD9401. In rat adipose tissue it stimulated hormone-sensitive lipase and inhibited acetyl-CoA carboxylase, as intact hGH does (Ng et al., 2000), and it raised lipolytic and lowered lipogenic activity in isolated adipose tissue from obese rodents and humans (Heffernan et al., 2000). Models: rodent and human adipose tissue in vitro.

The rodent studies cited here used AOD-9604 itself. In obese Zucker rats given 500 micrograms per kilogram by mouth daily for 19 days, body weight gain was under half that of controls, adipose lipolytic activity rose, and euglycemic clamps showed no adverse effect on insulin sensitivity, unlike chronic exposure to intact hGH (Ng et al., 2000).

In obese mice on 14-day osmotic pumps it raised fat oxidation and plasma glycerol without hyperglycemia, and it neither competed for the hGH receptor nor induced proliferation in cells carrying it (Heffernan et al., 2001). In beta-3 adrenergic receptor knockout mice the body weight and lipolysis changes seen in wild-type mice did not occur; the authors concluded that it raises beta-3 receptor expression but that its lipolytic action is not mediated directly through that receptor (Heffernan et al., 2001).

Phase IIa trials of AOD-9604 as an investigational obesity drug were under way by February 2002 (Wilding, 2004). A 2026 review reports that the oral candidate showed no significant effect on the primary body weight endpoint of a 24-week study with 502 participants randomized, based mainly on sponsor-reported outcomes; it cites the developer's 2007 announcement terminating the program and lists no reported human half-life and no meaningful IGF-1 elevation (Dominikowski et al., 2026). Model: human phase 2 trials.

No major regulator has approved AOD-9604 for any indication (Dominikowski et al., 2026). An FDA page on bulk drug substances that may present significant safety risks lists it among withdrawn compounding nominations formerly in category 2, citing a possible significant risk of immunogenicity for certain routes of administration, complexities with peptide-related impurities and API characterization, too little information to know whether it would cause harm in humans, and serious adverse events that may be associated with it, though causality is not clear.

Research Context: Tesamorelin

As TH9507, tesamorelin raised plasma GH and IGF-1 in pigs, rats and dogs. Its apparent elimination half-life in dogs was 21 to 45 minutes, and reversible adverse findings in dogs were attributed to sustained supraphysiological GH and IGF-1 (Ferdinandi et al., 2007).

In 13 healthy men over two weeks of once-daily exposure, it raised mean overnight GH, GH peak area, basal secretion and IGF-I, while fasting glucose and insulin-stimulated glucose uptake did not change significantly (Stanley et al., 2011). A 2026 review lists a human half-life of about 26 to 38 minutes (Dominikowski et al., 2026); current FDA labeling for Egrifta WR gives 11 minutes in healthy subjects after one subcutaneous exposure (Egrifta WR label).

In a randomized, placebo-controlled trial of 412 adults with HIV and abdominal fat accumulation (NCT00123253, registered as phase 3), visceral adipose tissue on CT fell 15.2% over 26 weeks with tesamorelin and rose 5.0% with placebo, while IGF-I rose 81.0% with tesamorelin and fell 5.0% with placebo. Glycemic measures and adverse events did not differ significantly, though more tesamorelin participants withdrew because of an adverse event (Falutz et al., 2007).

In November 2010 the FDA approved a pharmaceutical tesamorelin product for one indication, reducing excess abdominal fat in HIV-associated lipodystrophy (Grunfeld et al., 2011). That approval covers that drug product only, not the research material sold here.

Choosing Between Them for a Research Question

Choose by where the question sits. GHRH receptor signaling, pituitary GH release, pulsatility and IGF-1 point to tesamorelin, which raised GH peak area, basal secretion and IGF-I in healthy men (Stanley et al., 2011). Adipose lipolysis independent of the GH receptor points to AOD-9604, whose rodent studies measured adipose lipolytic activity, plasma glycerol and fat oxidation (Ng et al., 2000; Heffernan et al., 2001).

Do not borrow findings across the pair. Even a matching endpoint came from different models: insulin sensitivity by euglycemic clamp was measured in obese Zucker rats for AOD-9604 (Ng et al., 2000) and in healthy men for tesamorelin (Stanley et al., 2011). A PubMed search on 12 October 2026 found no study testing the two together, only reviews naming both. A head-to-head design needs a vehicle arm and IGF-1 measured in every arm.

Plan arms in moles: one milligram holds about 0.55 µmol of AOD-9604 and 0.19 µmol of tesamorelin. The Tesamorelin & Ipamorelin blend adds a ghrelin receptor agonist, so it cannot isolate the GHRH route.

Handling and Storage

All three ship lyophilized, and their product pages list -20°C for the powder as shipped and 2 to 8°C after reconstitution, protected from light, with freeze/thaw cycles avoided. Let the sealed vial reach room temperature before opening, add diluent slowly down the vial wall, and swirl rather than shake.

AOD-9604's disulfide is broken by reducing agents, so keep dithiothreitol and 2-mercaptoethanol out of its stock unless the experiment calls for them. Tesamorelin can cloud or gel in near-neutral, unbuffered diluents and at high concentration (see our tesamorelin gelling note), and its methionine 27 oxidizes readily. Aliquot single-use volumes, record the lot number, and use the measured content from the COA portal, not the label mass.

Questions Researchers Ask

AOD-9604 vs tesamorelin: what is the difference?

Class and site of action. AOD-9604 is a 16-residue analog of the C-terminal end of growth hormone that did not compete for the GH receptor (Heffernan et al., 2001); tesamorelin is a 44-residue GHRH analog that raised GH and IGF-I in healthy men (Stanley et al., 2011).

Tesamorelin vs AOD-9604: which has more human research?

Tesamorelin. A 2026 review places it in tier A, with randomized trials within an approved indication, and AOD-9604 in tier B/C, with short-term human trials and limited interpretable signals (Dominikowski et al., 2026).

Does AOD-9604 raise IGF-1 like tesamorelin?

Not in human data so far. Tesamorelin raised IGF-I by 81.0% in a phase 3 trial (Falutz et al., 2007); the 2026 review reports no meaningful IGF-1 elevation with AOD-9604 (Dominikowski et al., 2026).

Is AOD-9604 or tesamorelin an approved drug?

Only tesamorelin, and only as a pharmaceutical product approved by the FDA in November 2010 for one HIV-related indication, not as research material (Grunfeld et al., 2011). No major regulator has approved AOD-9604 for any indication (Dominikowski et al., 2026).

Are AOD-9604 and tesamorelin prohibited in sport?

Yes. The 2026 WADA Prohibited List names AOD-9604 as a growth hormone fragment under S2.2.3 and tesamorelin among GHRH analogues under S2.2.4, both in class S2, which is prohibited at all times.
