Research Overview
Kisspeptin and human chorionic gonadotropin (hCG) sit at opposite ends of the hypothalamic-pituitary-gonadal axis. Kisspeptins, peptides derived from the KISS1 gene product, activate the G protein-coupled receptor GPR54, now called KISS1R (Kotani et al., 2001), and a key action is at the level of GnRH release: in sheep, kisspeptin delivered into the brain ventricles released gonadotropin-releasing hormone (GnRH) into cerebrospinal fluid with a parallel rise in serum luteinizing hormone (LH) (Messager et al., 2005). hCG is a placental glycoprotein hormone that acts on the gonads through a receptor it shares with LH (Cole, 2010).
Kisspeptin vs hCG is therefore a comparison of the top and the bottom of one axis: a signal whose gonadotropin effect runs through GnRH neurons and the pituitary, against one that bypasses both. Disguised Alpha supplies Kisspeptin as kisspeptin-10 and hCG. Neither product name carries a strength: the 10 mg and 5000IU figures appear only in the certificate summaries and on the vials in the product photos. Both are lyophilized powders for laboratory research use only.
Structure and Class
Kisspeptins share an amidated RF C-terminus. Peptides of 54, 14 and 13 residues isolated from human placenta bound rat and human GPR54 with low nanomolar affinity in Chinese hamster ovary cells and triggered phosphoinositide hydrolysis and calcium mobilization (Kotani et al., 2001). Kisspeptin-10, the C-terminal decapeptide, is Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH2 (PubChem CID 25240297), C63H83N17O14 and 1302.4 g/mol, the figures on the product page.
hCG is a heterodimeric glycoprotein: a 92-residue alpha subunit almost identical to that of human LH and a 145-residue beta subunit, with sugar chains at two alpha and six beta positions (Morgan et al., 1975). Its molecular weight is about 36,000, 25 to 30% of it sugar in regular hCG (Cole, 2010); the intact hormone has no single formula or PubChem compound record (CID), and preparations are usually labeled in units of activity (IU).
Both receptors are G protein-coupled. In adult male rats, 77% of GnRH neurons co-expressed KISS1R messenger RNA (Irwig et al., 2004). The rat luteal LH/hCG receptor (LHCGR) joins a 341-residue leucine-rich extracellular domain to a 333-residue seven-transmembrane region; engineered cells expressing it bound hCG with high affinity and made more cyclic AMP, and its messenger RNA is prominent in the rat ovary (McFarland et al., 1989).

Research Context: Kisspeptin
Genetics placed kisspeptin signaling near the top of the axis. Affected members of a consanguineous family with idiopathic hypogonadotropic hypogonadism carried a homozygous L148S change in GPR54, and Gpr54-deficient mice showed isolated hypogonadotropic hypogonadism yet responded to exogenous gonadotropins and GnRH (Seminara et al., 2003). Models: a human pedigree and knockout mice.
In adult male rats, kisspeptin-52 delivered into the brain ventricles raised serum LH and induced Fos in 86% of GnRH neurons, and a GnRH antagonist given beforehand blocked the LH rise (Irwig et al., 2004); in mice lacking Gpr54, kisspeptin did not release LH or FSH (Messager et al., 2005).
In agonadal juvenile male monkeys, continuous kisspeptin-10 gave a brisk LH response for about 3 hours, then a precipitous drop; a later kisspeptin bolus failed while GnRH still released LH, which the authors read as desensitization (Seminara et al., 2006). Models: rats, mice and rhesus monkeys.
In a double-blind, placebo-controlled crossover study in six healthy men, a 90-minute kisspeptin-54 infusion raised plasma LH and FSH against saline, with a plasma half-life of 27.6 minutes (Dhillo et al., 2005). In a single-blind, placebo-controlled study in healthy men, kisspeptin-10, kisspeptin-54 and GnRH were each infused at three rates: the two isoforms gave similar gonadotropin secretion, and LH and FSH were about 3-fold higher with GnRH than with kisspeptin-10 and about 2-fold higher than with kisspeptin-54 (Jayasena et al., 2015). Model: healthy men.
Research Context: hCG
In male rats, a low amount of hCG raised testicular LH receptor numbers slightly before they fell to about 30% of control; a 20-fold larger amount caused faster, more complete loss, inhibited the cyclic AMP response for up to 5 days and blocked the testosterone response to hCG in vitro for about 3 days (Hsueh et al., 1977). Model: male rats.
After a rapid intravenous bolus of purified hCG in healthy young men and women, the disappearance curve ran over 8 to 10 days, with rapid and slow half-lives of about 6 and 36 hours (Wehmann and Nisula, 1981). Model: healthy adult volunteers.
The FDA list of applications deemed to be biologics licenses on March 23, 2020 includes five chorionic gonadotropin drug products first approved between 1973 and 1976, plus choriogonadotropin alfa, first approved in 2000. That history covers those drug products only; Disguised Alpha's hCG is research material, not those products.
Choosing Between Them for a Research Question
For kisspeptin vs hCG, start with the level of the axis. Kisspeptin questions concern KISS1R signaling, GnRH neuron activation and GnRH release, and its LH readout depends on GnRH: a GnRH antagonist blocked it in rats (Irwig et al., 2004). hCG questions concern LHCGR signaling and gonadal output, with no hypothalamic or pituitary step (McFarland et al., 1989).
Then match the time course. Kisspeptin-54's plasma half-life was 27.6 minutes (Dhillo et al., 2005) and continuous kisspeptin-10 lost its effect within hours in monkeys (Seminara et al., 2006), so kisspeptin work samples over minutes to hours and the exposure pattern is itself a variable. hCG half-lives of about 6 and 36 hours (Wehmann and Nisula, 1981) mean sampling over days, and in male rats hCG lowered testicular LH receptor numbers for days (Hsueh et al., 1977), so receptor number changes across that window.
Finally, read each at its own level: kisspeptin as GnRH release where it can be sampled, as in sheep cerebrospinal fluid (Messager et al., 2005), or as the LH and FSH that follow; hCG at the gonad, as receptor number or steroid output (Hsueh et al., 1977). Kisspeptin-10 is quantified by mass or moles and hCG in IU, so the two cannot be compared milligram for milligram.
Handling and Storage
Both ship lyophilized, and both product pages list -20°C for the powder as shipped and 2 to 8°C after reconstitution, protected from light, with freeze/thaw cycles avoided. Let a sealed vial reach room temperature before opening, add diluent slowly down the wall, swirl rather than shake, and split stock into single-use aliquots.
Record the lot number and work from the measured content in the COA portal: the July 2026 certificate for kisspeptin lot KS02 reports 10.49 mg against a 10 mg label, and the June 2026 hCG certificate (batch 35680) reports measured IU against the 5000IU label figure.
Questions Researchers Ask
Kisspeptin vs hCG: what is the difference?
Kisspeptin is a family of RF-amide peptides, supplied here as kisspeptin-10, that act on KISS1R at GnRH neurons (Irwig et al., 2004). hCG is a heterodimeric glycoprotein that acts at the gonad through the receptor it shares with LH (Cole, 2010).
hCG vs kisspeptin: which one needs a working pituitary?
Kisspeptin. Its LH response runs through GnRH and the pituitary, and a GnRH antagonist blocked it in rats (Irwig et al., 2004). hCG binds LHCGR on the gonad directly (McFarland et al., 1989).
Is hCG a peptide like kisspeptin?
Not strictly. Kisspeptin-10 has ten residues; hCG's two chains have 92 and 145 and carry eight sugar chains (Morgan et al., 1975), 25 to 30% of the weight of regular hCG (Cole, 2010). hCG is a glycoprotein; the hCG research guide covers its structure.
Are kisspeptin and hCG prohibited in sport?
Yes, for male athletes. The 2026 WADA Prohibited List names chorionic gonadotrophin (CG) and "kisspeptin and its agonist analogues" in section S2.2.1, under peptide hormones and their releasing factors. That section applies to males, and class S2 is prohibited at all times, in and out of competition.
Kisspeptin and hCG are supplied strictly for laboratory and in-vitro research use. They are not for human consumption, veterinary use, or any diagnostic or therapeutic application. Nothing on this page is guidance for use in people.
