Gonadorelin vs Kisspeptin
Research Comparison

A Research Use Only comparison of gonadorelin, the synthetic decapeptide GnRH that acts on GnRH receptors in the pituitary, and kisspeptin, the KISS1-derived peptides that act one step higher, mainly on KISS1R at the GnRH neurons of the hypothalamus.

Research Overview

Gonadorelin and kisspeptin act on neighboring steps of the hypothalamic-pituitary-gonadal axis. Gonadorelin is synthetic gonadotropin-releasing hormone (GnRH), the hypothalamic decapeptide that releases luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary (Schally et al., 1971). Kisspeptins, peptides derived from KISS1, activate GPR54, now called KISS1R (Kotani et al., 2001), and a key action sits one step up, at GnRH release: in sheep, kisspeptin delivered into the brain ventricles released GnRH into cerebrospinal fluid with a parallel rise in serum LH (Messager et al., 2005).

Gonadorelin vs kisspeptin is therefore a question of which step a study enters: the pituitary GnRH receptor, or the GnRH neuron that drives it; gonadorelin vs hCG and kisspeptin vs hCG cover the gonadal end. Disguised Alpha supplies Kisspeptin as kisspeptin-10, a lyophilized powder for laboratory research use only; the ILS certificates on its product page show vials labeled 10 mg. It does not list gonadorelin. Nothing on this page is guidance for use in people.

Structure and Class

Gonadorelin is pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, isolated from porcine hypothalami and confirmed by synthesis (Schally et al., 1971); PubChem CID 638793 lists the free peptide at 1182.3 g/mol, and its salts weigh more. Its receptor, cloned from mouse, is a 327-residue G protein-coupled receptor without a typical intracellular C-terminal tail, with a characteristic distribution in the rat anterior pituitary (Tsutsumi et al., 1992).

Kisspeptins share an amidated RF C-terminus: placental peptides of 54, 14 and 13 residues bound rat and human GPR54, a G protein-coupled receptor, with low nanomolar affinity in Chinese hamster ovary cells and stimulated phosphoinositide hydrolysis and calcium mobilization (Kotani et al., 2001). Kisspeptin-10, the C-terminal decapeptide (residues 112 to 121 of the parent protein; Chan et al., 2011), is Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH2 (PubChem CID 25240297): C63H83N17O14 and 1302.4 g/mol, as on the product page.

Research Context: Gonadorelin

As the hormone itself, gonadorelin is read at the pituitary as released LH and FSH, and its delivery pattern matters. In rhesus monkeys whose hypothalamic lesions abolished gonadotropin release, hourly GnRH pulses reestablished gonadotropin secretion and a constant infusion did not; continuous infusion in restored animals could desensitize release (Belchetz et al., 1978).

The peptide can also be measured in plasma. In healthy volunteers, a radioimmunoassay that does not detect GnRH fragments gave a first-phase half-disappearance time of 5.5 to 8 minutes after a constant infusion (Pimstone et al., 1977).

Drugs@FDA lists two US human drug applications for gonadorelin salts: NDA 018123, Factrel (gonadorelin hydrochloride), original approval 09/30/1982, and NDA 019687, Lutrepulse Kit (gonadorelin acetate), original approval 10/10/1989. Every product on both has the marketing status "Discontinued" (FDA, NDA 018123; FDA, NDA 019687). Separately, the Code of Federal Regulations lists gonadorelin solutions with conditions of use in cattle, restricted to use by or on the order of a licensed veterinarian (21 CFR 522.1077).

Research Context: Kisspeptin

Several lines of evidence place kisspeptin's main action at the GnRH neuron. In adult male rats, 77% of GnRH neurons expressed GPR54 messenger RNA; kisspeptin-52 delivered into the lateral ventricles raised serum LH and induced Fos in 86% of GnRH neurons, and the GnRH antagonist acyline, given beforehand, blocked the LH rise (Irwig et al., 2004).

Kisspeptin-10 released LHRH from rat hypothalamic explants but at 100 to 1000 nM did not change LH or FSH release from anterior pituitary fragments (Thompson et al., 2004), and kisspeptin had no effect on LH or FSH in mice lacking GPR54 (Messager et al., 2005).

The pituitary is not ruled out. In rats, systemically delivered KiSS-1 peptide raised LH to a maximum similar to central delivery, which the authors suggested might be partly a direct action at the pituitary (Navarro et al., 2005), and human GPR54 was highly expressed in the pituitary, among other tissues (Kotani et al., 2001).

In 13 healthy men, one kisspeptin-10 bolus produced an immediate LH pulse whatever the timing of the previous one and delayed the next by about one normal interpulse interval. From pulse shapes compared with GnRH-induced LH pulses in men with isolated GnRH deficiency, the authors estimated about 17 minutes of GnRH release and called kisspeptin the first known agent able to reset the GnRH pulse generator (Chan et al., 2011). A search of Drugs@FDA found no kisspeptin or metastin product (October 2026).

Choosing Between Them for a Research Question

For kisspeptin vs gonadorelin, start with the step of the axis. Gonadorelin fits questions about GnRH receptor signaling, gonadotrope desensitization or the pituitary response to a defined GnRH pattern. Kisspeptin fits questions about KISS1R signaling, GnRH neuron activation or pulse generation, with an LH readout that depends on GnRH release (Irwig et al., 2004).

Studies that separate the two steps use both. In three agonadal juvenile male monkeys, continuous kisspeptin-10 gave a brisk LH response for about 3 hours, then a precipitous drop; on day 4 a kisspeptin bolus no longer released LH while NMDA and GnRH boluses still did, which the authors read as GPR54 desensitization with GnRH neurons and receptors intact (Seminara et al., 2006). Continuous GnRH, by contrast, can desensitize release at the pituitary (Belchetz et al., 1978).

Then read the output at the right level. With gonadorelin, LH is a direct pituitary response; with kisspeptin, it is a proxy for GnRH release, which can be sampled directly only in some models, such as sheep cerebrospinal fluid (Messager et al., 2005).

In a single-blind, placebo-controlled study in healthy men, five per group, kisspeptin-10, kisspeptin-54 and GnRH were each infused for 3 hours at the same three molar rates. Serum LH and FSH were about 3-fold higher with GnRH than with kisspeptin-10 and about 2-fold higher than with kisspeptin-54, the isoforms were similar at the rates tested, and the authors noted the small sample size (Jayasena et al., 2015).

Finally, keep the units straight. One milligram holds about 0.85 µmol of gonadorelin free peptide and about 0.77 µmol of kisspeptin-10, so equal masses are not equal amounts of peptide; the 2015 comparison matched the hormones by moles.

Handling and Storage

Kisspeptin ships lyophilized, and its product page lists -20°C for the powder as shipped and 2 to 8°C after reconstitution, protected from light, with freeze/thaw cycles avoided. Bring the sealed vial to room temperature before opening, swirl rather than shake, and aliquot. Record the lot number and use the measured content from the COA portal: the July 2026 ILS certificate for lot KS02 reports 10.49 mg against a 10 mg label.

Protease cleavage of kisspeptin-10 and the solution stability of gonadorelin by pH are covered in the kisspeptin and gonadorelin research guides.

Questions Researchers Ask

Gonadorelin vs kisspeptin: what is the difference?

Gonadorelin is synthetic GnRH, a ten-residue peptide that releases LH and FSH through pituitary GnRH receptors (Schally et al., 1971). Kisspeptin acts mainly one step higher, on KISS1R on GnRH neurons, and in male rats its LH rise after central delivery needed GnRH release (Irwig et al., 2004). They belong to different peptide families.

Kisspeptin vs gonadorelin: does kisspeptin act on the pituitary directly?

Its main action is on GnRH neurons. Rat anterior pituitary fragments did not respond to 100 to 1000 nM kisspeptin-10 (Thompson et al., 2004), but one rat study read part of the LH response to systemic KiSS-1 peptide as a direct pituitary action (Navarro et al., 2005). Gonadorelin acts on the pituitary GnRH receptor itself.

Are gonadorelin and kisspeptin prohibited in sport?

Yes, for male athletes. The 2026 WADA Prohibited List names gonadotrophin-releasing hormone (GnRH, gonadorelin) and its agonist analogues, and kisspeptin and its agonist analogues, in section S2.2.1, under peptide hormones and their releasing factors. That section applies to males, and class S2 is prohibited at all times, in and out of competition.

Does Disguised Alpha sell gonadorelin?

Not currently. Disguised Alpha supplies Kisspeptin (kisspeptin-10) for laboratory research.
